Will Zorin 16 beta upgrade to Zorin 16 automatically through software-update? or I need to upgrade manually?
I think you can upgrade to Zorin Os 16 from beta but the developers recommend you to do a clean install.

Yes, you are right. It is possible, but best practice is to do clean install.
You can update and have no issues ,but can be opposite also ( you can have lot of issues).
New install is just 10 min 
Plus the time required for restoring the customization 
I know, that is the pain always. But maybe someone likes default setup, than it is 10 min xD
